段金伟,2014年7月于兰州大学化学化工学院物理化学专业取得理学博士学位落伍入强强彩票App事情,次要从事DNA/RNA纳米构造的拓扑剖析及模仿设计、实行分解和使用研讨,在Topics Curr. Chem.,Biochem. Bioph. Res. Co., J. Mol. Graph. Model., MATCH Commun. Math. Comput. Chem., J. Mol. Chem.等国际期刊上揭晓学术论文17篇,掌管并完成陕西省天然迷信基金面上项目和青年项目各1项,掌管和完成地方高校根本研讨项目2项,掌管和介入教改项目2项。2019年在国度留学基金委赞助下前去美国Rensselaer polytechnic institute(RPI)和University of Illinois--Urbana-Champaign(UIUC)访学1年,与Wang Xing传授互助展开DNA纳米构造的分解及使用研讨,经过培育癌细胞,培育细菌提取plasmids,对培育细胞举行转染,经过荧鲜明微镜检测细胞熏染情形,实验为基因交融驱动的癌症患者提供特性化的医治方案。

Chromosome fusion events are endemic to many cancers and in many cases are known drivers of oncogenic transformation. An accurate assessment of any chromosomal fusion’s role in cellular proliferation and survival can be established in vitro by breaking apart the fusion gene by CRISPR and comparing growth and viability relative to untreated cancer cells. In order to explore the therapeutic application of the CRISPR to reverse chromosome fusions in vivo, we innovate in molecular targeting specificity by using a deactivated Cas9 (dCas9) fused with a FokI nuclease to ensure that DNA cleavage will occur only in cancer cells and leave the healthy genome unaffected and then we propose an innovative DNA origami nanostructure (DNA hollow sphere)-based cell targeting and delivery system to enable the delivery of the CRISPR complex specifically to cancer cells. The ability to prove that a given fusion is an oncologic driver is important to understanding the pathology of the disease and our ‘personalized medicine’ approach to cancer therapy advances proposed will be broadly transformative and significant.
